Tsukune Aono is a normal high school teenager, who is unable to get into any private school due to his poor grades. However, with the help from his parents, he gets enrolled to a private school called Yokai Academy. When he arrives at the school, he soon discovers that Yokai Academy is a secret private school for supernatural monsters and mythological creatures disguised as humans, and any human who finds out about its existence will eventually be killed without mercy. Realizing this problem, Tsukune fears for his life, thinking that his admission into Yokai Academy is a mistake. His luck soon changes when he quickly becomes friends with a beautiful girl Moka Akashiya, who turns out to be a vampire, attracted to the sweet taste of his blood, sometimes biting Tsukune on the neck when the chance seems right. While at the beginning of the series, Moka was upset upon discovering Tsukune's human identity, as she realizes that he is the first real friend she had ever made in her life. Despite Moka's vampiric nature, she is a kind girl and Tsukune quickly falls for her, and ultimately decides to stay at Yokai Academy, despite the various dangers while hiding his human identity from the faculty and other students. Tsukune soon discovers that when he removes the rosario around Moka's neck, she transforms into her true vampire form (her true perspective and personality is sealed, the sweet Moka being a different version or an artificial person), being stronger and more powerful than almost any other creature at the school, and Tsukune is the only person who can physically remove it in order for her to transform into her true self. However, "Inner Moka" has an entirely different personality, often cold and merciless, and thus the rosary is only removed in dire emergencies.
As the school year continues, Tsukune becomes involved with other students who start out as enemies, but who are won over into becoming his friends: Kurumu Kurono, a busty succubus; Ginei Morioka, a perverted lech of a werewolf who is a second-year student and president of the school's Newspaper Club; Ruby Tojo, a witch from the human world, Yukari Sendo an eleven-year-old witch; and Mizore Shirayuki, a yuki-onna (a teen version of the legendary snow woman of Japanese folk tales) who stalks Tsukune, believing them to be soulmates. However, after Tsukune befriends each of the girls, they become friendly rivals along with Moka, vying for Tsukune's affections, and later most become members of the school's Newspaper Club with Tsukune and stand by him even after they learn that he is really a human. Together they wind up fighting against a variety of enemies within the school body, be it students or teachers, who try to use their monstrous powers to bully or cowtow one or more of the group for their own selfish or destructive ends.
Later on in the second manga serialization, Kokoa Shuzen, Moka's younger half-sister, appears. And, as time goes on, they discover dangerous organizations like the Anti-Schoolers and Fairy Tale, and must do battle against them to keep the school and both the monster and human worlds safe; and Tsukune starts to undergo changes that will truly solidify his role as the link between the human and monster worlds.
Eventually it is revealed the Three Dark Lords defeated a powerful monster named Alucard, which tried to destroy the world; it shared the same bloodline as Moka and her mother. The rosario is also revealed to seal Moka's true personality away, meaning Outer Moka is artificial; the organization known as Fairy Tale kidnaps Moka as well. During the Newspaper club's attack on Fairy Tale HQ, the rosario's final secret is revealed; it's meant to control Alucard if he ever awakens. As of now, Fairy Tale controls the most dangerous being in existence.

La historia se concentraba en mantener el secreto de Tsukune, el cual era su verdadera forma, un ser humano, ya que en la Academia Youkai ningun humano es bienvenido. Pero, esto cambia cuando este es descubierto por los de la comisión de seguridad publica, arriesgando su vida para salvar a Inner Moka (Moka interna), donde el muere, pero por la inyeccion de sangre de esta el se salva. Todo da un giro con la transformación de Tsukune en ghoul debido a las múltiples veces que Moka inyectó sangre en él para salvarlo y por la amenaza de Outcast Ayashi de matarlo. A esto hay que agregar la llegada y posterior batalla con el grupo “Antitesis”, que desea unir el mundo de los monstruos con el de los humanos, para que así haya un caos total. La batalla con el líder de Antitesis, Hokuto, y el posterior desastre causado por el Espejo de Lilith, marcan el final del primer arco argumental.
La historia continua con la secuela llamada Rosario + Vampire Season II, que se enfoca más en la familia de Moka y la explicación de su oscuro y olvidado pasado. Es importante destacar la adición del personaje de Kokoa, quien es la hermana menor de Moka.

Tsukune Aono est un jeune garçon, tout à fait banal, qui,
malheureusement, a raté tous les examens d'entrée au lycée. Par un pur
hasard, il est alors relégué dans un établissement à première vue banal,
mais qui n'est en fait ni plus ni moins qu'un lycée… pour monstres !
Là, il fait la rencontre de Moka Akashiya, jeune fille, à l'apparence
angélique, qui cache en réalité une double personnalité et des pouvoirs
de vampire
très puissants qui se révèlent lorsque l'on arrache le rosaire ornant
son cou. Grâce aux pouvoirs de Moka, tout deux vont combattre plusieurs
monstres. Plus tard, Tsukune et Moka rencontreront la succube Kurumu Kurono et Ginei Morioka, président du club de journalisme, un loup-garou
surnommé « Gin ». Dans le deuxième tome, on découvre aussi Yukari
Sendo, une petite sorcière surdouée. Et enfin par la suite, ils
rencontreront Rubi Tōjō,une autre puissante sorcière, et Mizore
Shirayuki (Mizoré phonétiquement) qui est une femme des glaces ayant
quelques tendances de rôdeuse (Stalker en japonais)...Italiano
Tsukune Aono è un normale studente di scuola superiore, che non è stato in grado di entrare in una scuola privata a causa dei suoi voti scadenti. L'unica scuola che finalmente lo accetta in realtà si rivela essere una scuola segreta per mostri soprannaturali e creature mitiche (yōkai), incarnati in esseri umani. Quando Tsukune viene ammesso nella scuola yōkai per errore, lui che ha ormai scoperto la vera natura di quel luogo, teme per la sua vita.
Comunque il ragazzo fa rapidamente amicizia con la bella Moka Akashiya, che si rivela essere un vampiro, attratta dal dolce gusto del sangue. Mentre in un primo momento Moka rimane sconvolta nello scoprire che Tsukune è un normale essere umano, si rende in seguito conto che lui è il primo vero amico che abbia mai avuto.
Nonostante la natura di vampira, Moka è una ragazza gentile e Tsukune
si innamora subito di lei così come Moka di lui. Tsukune decide quindi
di rimanere vicino a lei, nonostante il pericolo. Per fare questo è
costretto a nascondere la sua natura umana alla scuola e agli altri
studenti per evitare conflitti con i vari mostri presenti nel campus.
Tsukune scopre presto che quando toglie il "rosario"
dal collo di Moka, lei si trasforma in un super-vampiro, più forte e
più potente rispetto a qualsiasi altra creatura della scuola. Tuttavia,
in questo stato, ha un carattere completamente diverso, diventa fredda e
spietata, quindi Tsukune, che è l'unico a poter fare la cosa, ricorre a
questo accorgimento solo in casi di estrema emergenza.L'anno scolastico continua, e Tsukune viene pian piano accolto dagli altri studenti che, inizialmente ostili, alla fine diventano suoi grandi amici: Kurumu Kurono, succubus, Yukari Sendo, strega e studentessa più intelligente dell'Accademia, Ginei Morioka, studente del secondo anno, presidente del Club del Giornale della scuola, e lupo mannaro e infine Mizore Shirayuki, yuki-onna (letteralmente donna delle nevi).
Col passare del tempo, si scopriranno nella scuola pericolose organizzazioni, come Antitesi e Fairy Tale, quindi occorrerà combattere per cercare di mantenere la scuola e il mondo umano sicuri.
Nel frattempo Tsukune inizia a subire delle trasformazioni per consolidare il suo ruolo di collegamento tra il mondo umano e mondo yōkai.
